Should I be running the exhaust and intake fans 24 hrs ?

Yes.

At least the exhaust . . . no air movement at all in a warm moist environment is an invitation for fungi. In low humidity I stop the circulation fan at night . . . high humidity I run everything, intake, exhaust and circulation fans 24/7.

yes, same here. I have 3 grow tents, each have oscillating fans and 4" exhaust fans, all running at level 9/10 to ensure negative pressure to ensure air is constantly flowing into the tent via the vents with air filters I add on them. If you have an intake fan, then you don’t need to create the negative pressure I believe.

All my inline fans feed into an exhaust line that also has a 6" inline fan at the end, where it exhausts outside. I do this in case any inline fan in a tent dies, the 6" fan will still move air, and also it ensures that one tent is not venting into other tents.

Everything but my lights run 24/7

Remember, the whole negative pressure idea is to control odor . . . a filtered intake and positive pressure helps prevent mold spores, insects, etc. as they aren’t pulled into the grow area at every crack or crevasse
